EC medium (or EC Broth, or Escherichia coli Broth) is a selective enrichment broth designed for the isolation of coliforms, including E. coli, from water and food samples using the Most Probable Number (MPN) method. It uses bile salts to inhibit gram-positive bacteria and uses elevated temperature to distinguish fecal coliforms from (Incubated at 44.5 °C for 24 hours) from coliforms (37 °C).

The addition of 4-methylumbelliferyl-β-D-glucuronide (MUG) allows rapid detection of E. coli. MUG is used as a substrate to detect glucuronidase activity. Its degradation by this enzyme produces the fluorescent compound 4-methylumbelliferone, easily visible under UV light.
